Brooke Greenberg is the size of an infant, with the mental capacity of a toddler.

She turned 16 in January.

Younger sister says "we joke about how she rebels" like a true 16-year-old.

"Why doesn't she age?" Howard Greenberg, 52, asked of his daughter. "Is she the fountain of youth?"

Such questions are why scientists are fascinated by Brooke. Among the many documented instances of children who fail to grow or develop in some way, Brooke's case may be unique, according to her doctor, Johns Hopkins School of Medicine pediatrician Lawrence Pakula, in Baltimore.

"Many of the best-known names in medicine, in their experience ... had not seen anyone who matched up to Brooke," Pakula said. "She is always a surprise."

Watch "20/20" FRIDAY at 10 p.m. ET for Brooke's story.

Brooke hasn't aged in the conventional sense. Dr. Richard Walker of the University of South Florida College of Medicine, in Tampa, says Brooke's body is not developing as a coordinated unit, but as independent parts that are out of sync. She has never been diagnosed with any known genetic syndrome or chromosomal abnormality that would help explain why.

In a recent paper for the journal "Mechanisms of Ageing and Development," Walker and his co-authors, who include Pakula and All Children's Hospital (St. Petersburg, Fla.) geneticist Maxine Sutcliffe chronicled a baffling range of inconsistencies in Brooke's aging process. She still has baby teeth at 16, for instance. And her bone age is estimated to be more like 10 years old.

"There've been very minimal changes in Brooke's brain," Walker said. "Various parts of her body, rather than all being at the same stage, seem to be disconnected."

Brooke's mother, Melanie Greenberg, 48, sees a different picture. "She loves to shop," Greenberg said. "Just like a woman."

Brooke rides in a stroller while her mom shops for clothes in the infant sections of department stores near their home in a Baltimore suburb. That Brooke is in her mid-teens is so mind-boggling that if another mother with a toddler asks Greenberg how old Brooke is, she usually doesn't try to explain.

"My system always has been to turn years into months," Greenberg said. "So, if someone asked today, I might say, she's 16 months old."

It's so fascinating. Almost like Benjamin Button.

Don't forget to watch "20/20" FRIDAY at 10 p.m. ET for Brooke's story.

I saw that. There is a similar case in India of a man who has never aged and is still like a baby.

Wow, that's pretty rare and I think she's a speical girl. It's good to know she has her loving and caring family. :)

Great. I'm curious because of her case, I wonder how long will she live as a baby.

Thanks for the link, Bottesini.

If doctors do manage to finally find out the cause of this, they might also figure out how to control aging.

Wow, that's something new to me....that's really scary at the same time.

This was a really interesting story. I have a very close friend who has a 6 year old child that is still almost the size of an infant. But she was born with 2 chromosomal deficiencies -- missing two of them.

Wow, that is very interesting story. I guess it will takes years for doctors/scientists to find a specific gene that control the aging, especially there are too many other genes in DNA (Deoxyribonucleic acid).

old news. I thought it was because of defective pituitary gland or something. An organ that's responsible for producing growth hormone.

:dunno:

Not in this case. It is not just a growth issue, but an issue of failure to age of all the body systems. Someone with a pituitary deficiency might not grow, but their body still ages.

There are only a handful of people who have been diagnosed with such a disorder all over the world. This girl's case is unique from all of those. They suspect a gene mutation is responsible, and are currently attempting to isolate that gene from her DNA.

Her mentality is still of an infant or toddler? as well?

Kind of confused about that part.

They featured this family on 20/20 last night.

Mentally they say she is like a 9 month old .

Yes. Her brain development has not advanced past the age of approximately 6-9 months. It is not just an issue of comprehension, as in a child that has developmental delays or traumatic brain injury. A child like that still shows aging in the tissues of the brain. The tissue of this child's brain has failed to age. Likewise with her bone age. Some of her organs have shown the aging process to some extent, but are still significantly below that which would be expected in a 16 year old.

Oh wow!

That is highly unusual!

I used to say "I wish my kids could stay little!"

This Medical Marvel has just defeated that saying.

Doctors say her particular case is one of a kind, because her body is aging as separate parts rather than as a whole. There have been some similar cases, but nothing that is exactly like her case. That is why they believe it is a gene mutation.

Her bones are at 10 years old. her mind is at 9 months.

Seems like her whole system is out of sync. I wonder how long she will live?

absolutely amazing!
