Using the word "queer" is acceptable if you embrace gay and lesbian culture. For that, you'd need to be GLBT yourself, or move in GLBT circles.

If you're using the word in a derogatory manner, it is unacceptable. To use it for empowerment (as the creators of Queer As Folk did), it is perfectly acceptable.
